Anti-angiogenesis effect of generation 4 polyamidoamine/vascular endothelial growth factor antisense oligodeoxynucleotide on breast cancer in vitro

Objective: To study the effects of the generation 4 polyamidoamine/vascular endothelial growth factor antisense oligodeoxynucleotide (G4PAMAMNEGFASODN) compound on the expressions of vascular endothelial growth factor (VEGF) and its mRNA of breast cancer cells and on the inhibition of vascular endothelial cells. Methods: We examined the morphology of G4PAMAM/VEGFASODN compound and its pH stability, in vitro transfection efficiency and toxicity, and the expressions of VEGF and its mRNA. Methyl thiazolyl tetrazolium assay was used to detect the inhibitory function of the compound on vascular endothelial cells. Results: The compound was about 10 nm in diameter and was homogeneously netlike. From pH 5 to 10, it showed quite a buffered ability. The 48-h transfection rate in the charge ratio of 1:40 was 98.76%, significantly higher than that of the liposome group (P<0.05). None of the transfection products showed obvious toxicity on the cells. The expressions of both VEGF protein and its mRNA after G4PAMAM/VEGFASODN transfection decreased markedly. Conclusion: With a low toxicity, high safety, and high transfection rate, G4PAMAMNEGFASODN could be a promising gene vector. Specifically, it inhibits VEGF gene expression efficiently, laying a basis for further in vivo animal studies.  相似文献

由于单核苷酸多态性(SNPs)在寻找致病基因，了解遗传多样性、环境与基因的相互作用方面有重大价值，SNPs研究已越来越多地引起了学术界和产业界的兴趣．CYP1A1编码的酶在烟草的主要几类前致癌物(如PAHs和芳香胺类物质)的代谢活化中有重要作用，其多态性与个体对环境相关的癌症的易感性密切相关．本研究采用PCR-RFLP技术在上海人群中，对CYP1Al的两个SNPs：CYP1A1 m1和m2进行了基因分型．结合其它已发表的数据，我们的研究表明，在中国人群的不同群体中，m1等位基因的频率分布比较一致，而m2等位基因的频率分布则存在着显著的不同．  相似文献   

“It’s My Job Now,I Guess”: Biographical disruption and communication work in supporters of young adult cancer survivors

ABSTRACT

This study uses narrative interviews to describe how 25 young adult (YA) close supporters (i.e., romantic partners, siblings) experience an often invisible and uniquely burdensome biographical disruption following a young adult cancer survivor’s (YACS; i.e., 18–39) diagnosis. YA supporters reported varying levels of biographical interdependence with the YACS and family system uncertainty with other important supporters. These factors led YA romantic partners and siblings to engage in different types of communication work to negotiate and maintain primary and secondary support roles with parents and in-laws, to ensure that the YACS received helpful social support by gatekeeping (i.e., correcting, blocking) others’ support attempts, and to accommodate both their own and the YACS’s disrupted biographies.  相似文献

洛铂联合重组人血管内皮抑素胸腔内注射治疗肺癌恶性胸腔积液临床研究

目的：研究洛铂联合重组人血管内皮抑素（恩度）治疗肺癌恶性胸腔积液的有效性和安全性。方法：对确诊为肺癌恶性胸腔积液的63例患者进行分析,采用胸腔内注射治疗方法,其中32例注入洛铂和恩度（A组）,31例注入顺铂和恩度（B组）,比较两组间疗效、生活质量和毒副作用。结果：洛铂和恩度组与顺铂和恩度组在疗效和生活质量改善上比较差异无统计学意义（P〉0．05）,在毒副作用方面,洛铂和恩度组白细胞（WBC）减少、贫血、恶心呕吐发生率较顺铂和恩度组低,差异有统计学意义（P〈0．05）。结论：洛铂联合恩度胸腔内注射治疗肺癌恶性胸腔积液疗效与顺铂联合恩度相当,毒副作用较轻。  相似文献

Objective:The aim of this study was to evaluate the association between the methylenetetrahydrofolate reductase(MTHFR) C677T excision repair cross-complementation group 1(ERCC1) genetic polymorphisms and the clinical efficacy of gemcitabine-based chemotherapy in advanced non-small cell lung cancer(NSCLC).Methods:A total of 135 chemonaive patients with unresectable advanced NSCLC were treated with gemcitabine/platinum regimens.The polymorphisms of MTHFR C677T,ERCC1 C8092A,and ERCC1 C118T were genotyped using the TaqMan methods.Results:The overall response rate was 28.9%.Patients with MTHFR CC genotype had a higher rate of objective response than patients with variant genotype(TT or CT)(41.2% versus 19.1%,P=0.01).Median time to progression(TTP) of patients with MTHFR CC genotype was longer than that of patients with variant genotype(7.6 months versus 5.0 months,P=0.003).No significant associations were obtained between ERCC1 C118T and C8092A polymorphisms and both response and survival.Conclusions:Our data suggest the value of MTHFR C677T polymorphism as a possible predictive marker of response and TTP in advanced NSCLC patients treated with gemcitabine/platinum.  相似文献

Experiences in public library cooperation have shown that the acceptance of cooperation as an ideal, patron reaction and acceptance, local pride and control, and the education and cooperation of the library board, local government officials, and funding agencies are all important in providing the best possible framework for cooperation. The commitment to and understanding of the implications, changes, and modifications that cooperation requires are also important. Audre Lorde's speech, “The Transformation of Silence into Language and Action, “sheds light on the margins of rhetoric in the sense of the public speech because she examines factors that may cause some people to remain silent while enabling others to speak and act. “Margins” refers both to the parameters employed for defining a practice and the relative place or value of varied activities exemplifying the practice. Lorde interweaves her commentary on the silence surrounding breast cancer with insights about silence drawn from her experiences as a member of several subordinated communities, especially as they relate to the misuse of power to silence those who are different. Her speech comments on silencing and power, sexism, verbal abuse, violence and sexualized aggression, shame, the taboo, and hostile social environments. Paradoxically, Lorde's speech is as much about the possibilities of rhetoric as its limits.  相似文献
